Wistar 乳鼠中脑黑质器官型脑片的培养
【目的】探索中脑黑质器官型脑片的培养方法, 寻找一种较好的帕金森病组织模型材料。【方法】选出生5 d ～ 7 d 的Wistar 乳鼠, 制备中脑黑质层面脑片。用Millicell-CM 微孔膜将脑片放置在培养液和气体界面上培养。分为培养0 d 、10 d 、20 d 和30 d 4 组, 进行倒置显微镜观察、酪氨酸羟化酶(TH)免疫组化和电镜超微结构观察。【结果】培养10 d 和20 d 的脑 片与0 d 对照组相比无明显差异。培养30 d 脑片发现有部分神经元变性坏死, 胶质细胞明显增生, TH 阳性神经元减少。同 时发现在脑片长期培养过程中存在“微血流” 现象。【结论】Milli...
