转γ -干扰素基因大鼠肝细胞模型的建立
目的: 使用多聚阳离子脂质体载体( lipofectAMINE)在体外建立转γ -干扰素( IFN-γ )基因的大鼠肝细胞模型。方 法: 利用 lipofectAMINE 在体外将 IFN-γ基因转染大鼠肝细胞。分别以 RT-PCR 和 ELISA 方法检测所转IFN-γ基因在大鼠肝细胞 的转录和表达情况。结果: 以 lipofectAMINE 为载体的 IFN-γ基因转染大鼠肝细胞后, 可被有效的转录并产生具有生物活性的 IFN-γ。结论: IFN-γ基因可被成功转染入大鼠肝细胞并可有效表达。这可能为细胞因子的免疫治疗提供一种新途径。...
