    上调microRNA-205表达对HeLa细胞生物学特性的影响

    “…【目的】 探讨上调microRNA-205(miR-205)表达对HeLa细胞生物学特性的影响65377;【方法】 实验分4组:miR-205上调组,阴性对照组,空白转染组和空白对照组65377;每种检测重复3次65377;利用脂质体siPORT NeoFX Transfection Agent,向miR-205上调组细胞转染Pre-miR-hsa-miR-205 miRNA Precursor,阴性对照组细胞转染Cy3 dye labeled PremiR Negative Control65377;荧光显微镜下评估转染情况,实时定量PCR方法检测miR-205表达,四甲基偶氮唑蓝(MTT)法和平板克隆形成实验分别检测细胞生长和增殖能力,流式细胞术检测细胞凋亡率和细胞周期分布,Transwell小室法检测细胞迁移能力65377;【结果】 实时定量PCR结果显示,miR-205上调组miR-205的表达增加3061.5倍65377;与空白对照组相比,72 h miR-205上调组与阴性对照组活细胞数分别为(126 ± 41)%,(126 ± 39)%;两组克隆形成率分别为(79 ± 11)%,(63 ± 10)%;凋亡率分别为(4.6 ± 2.1)%,(4.1 ± 2.4)%;S期细胞比例分别为(19.2 ± 3.6)%,(23.4 ± 3.0)%;200倍镜视野下迁移细胞数分别为63 ± 17和68 ± 1665377;与对照组比,miR-205上调组细胞增殖能力增强但生长曲线不受影响,S期细胞比例降低,而凋亡率不变,细胞迁移能力无明显改变65377; 【结论】 上调miR-205表达后,HeLa细胞增殖能力及细胞周期改变,但生长曲线65380;凋亡率和迁移能力无变化65377;…”
