“…【目的】 观察创伤性休克大鼠肝组织肿瘤坏死因子-α(TNF-α) mRNA表达65380;肝功能和病理改变的动态变化及甘氨酸的干预影响65377;【方法】 建立创伤性休克动物模型,110只Wistar大鼠随机分成3组:治疗组(n=40)65380;休克组(n=40)和对照组(n=30)65377;治疗组大鼠在复苏开始时经颈静脉输入100 mg/kg的甘氨酸液;休克组输以同体积的生理盐水;对照组仅行腹腔麻醉65377;分别于复苏后3 h65380;6 h65380;12 h65380;24 h及48 h五个时相点活杀大鼠65377;采用RT-PCR法检测肝组织TNF-α mRNA表达;观察肝组织病理改变并测定血清谷丙转氨酶(ALT)和谷草转氨酶(AST)水平65377;【结果】 休克组复苏后3 h肝组织TNF-α mRNA表达即增加,复苏后6 h达高峰65377;血清ALT65380;AST在复苏后3 h有升高,且随时间延长逐渐增高;光镜下病理改变也随时间延长而呈损害加重趋势65377;与休克组比较,治疗组各时间点肝组织TNF-α mRNA表达明显降低(P < 0.05)65380;血清ALT和AST明显降低(P < 0.05)且肝组织光镜下病理损害明显改善(P < 0.05)65377;【结论】 创伤性休克后肝组织TNF-α表达增加;甘氨酸可降低肝组织TNF-α表达,减轻创伤性休克后继发的肝脏损害65377;…”
