短时间周期性母婴分离与慢性疼痛对幼鼠空间学习记忆的影响

“…【目的】 研究短时间周期性母婴分离65380;弗氏完全佐剂(CFA)引起的慢性疼痛以及两者合并对幼鼠空间学习记忆行为影响65377;【方法】 20窝新生SD大鼠,10窝无母婴分离,每窝随机取两只分别作为空白对照组(C组)和疼痛组(P组),另外10窝进行母婴分离,每窝随机取2只分别作为母婴分离组(MS组)和母婴分离合并疼痛组(MS + P组)(n=10)65377;在幼鼠出生后第2天(D2),P组左足底皮下注射CFA 20 μL;C组注射0.9%生理盐水20 μL;MS组左足底皮下注射0.9%生理盐水20 μL后每天将幼鼠与母鼠分离10 min,从D2持续至D21;MS + P组左足底皮下注射20 μL CFA,随后处理同MS组65377;检测幼鼠在D2和D21的体质量变化;幼鼠从D22至D29进行Morris水迷宫实验(n=10)65377;【结果】 4组幼鼠体重增加类似,差异无统计学意义65377;Morris水迷宫实验中,P组寻找隐蔽平台的潜伏期比C组延长25.38% [(32.75 ± 2.71)s vs ( 24.28 ± 1.37)s, P < 0.05],比MS组(22.92±1.83)s 延长47.72%(P < 0.01); MS+P组的潜伏期(25.34±3.23)s 比P组减少22.61%(P <0.05);MS组65380;MS+P组与C 组的潜伏期无统计学意义;空间探索实验中,各组幼鼠穿过平台所在象限的时间占总游泳时间的比例以及穿过平台所在象限的路径占总路径的比例的差异均有统计学意义(F = 4.28, P < 0.05; F = 3.75,P < 0.05)65377; P组的时间比例和路径比例比C组和MS组减少,而MS + P组比P组延长65377; 【结论】 CFA诱导的慢性疼痛损害了幼鼠的空间学习记忆能力,而短时间周期性的母婴分离可以逆转慢性疼痛对幼鼠空间学习记忆能力的损害65377;…”
