Effect of Jiangu Granule on miR-141 and Osteogenic Gene Dlx5/Msx2/Runx2 in Ovariectomized Mice
Published 2018-12-01“…Fat around ovary of the sham operation group was removed, but not ovary, bilateral ovaries of the saline group and Jiangu granule group were removed surgically, and administrate respectively by normal saline and Jiangu granule.After eight weeks of intervention, the tibial morphology was observed by micro-CT, the expression of microRNA-141, Dlx5, Msx2 and Runx2 in bone tissue were detected by real-time PCR, protein contents of Dlx5, Msx2 and Runx2 in bone tissue were detected by Western blot.Results:Compared with the saline group, the number, width, length, shape and distribution of trabeculae in the Jiangu granule group recovered partially, and the density and connectivity of trabeculae increased, which could not be restored to the level of the sham operation group, The area of cortical bone increased, the bone marrow cavity decreased, and basically restored to the level of the sham operation group; BMC, BMD, BV/TV, BS/TV, Tb.N increased significantly; Tb.Sp, SMI reduced significantly;the relative amount of microRNA-141 and Msx2 in bone tissue decreased significantly;the relative contents of Runx2 and Dlx5 in bone tissue increased significantly, with statistically significant differences (<italic>P</italic><0.05).Conclusion:Jiangu granule can effectively reduce the expression of miR-141, enhance the expression of key osteogenic factors such as Dlx5 and Runx2 by influencing BMP signaling pathway Dlx5/Msx2/Runx2 in bone marrow mesenchymal stem cells (BMSCs), and promote osteogenic potential, so as to prevent and treat osteoporosis.…”
