A Method and Engineering Practice for a Fully Mechanized Caving Coalface to Rapidly Pass Through a Large Fault by Wei Zhang, Feili Yang, Jingyu Chang, Shengxun Zhao, Bin Xu, Jinyong Xiang

    Published 2025-01-01
“…The main implementation steps of the method include designing the layout parameters of the pre-driven roadways, determining the width for rock pillars between the adjacent pre-driven roadways, construction of pre-driven roadways by smooth wall blasting, and controlling the surrounding rock deformation of the pre-driven roadways. The results of engineering practice show that it took only 23 days for this coalface to pass through fault F6, about one month shorter than the time required by traditional methods (e.g., proactively taking a detour). …”

Overcoming CD226-related immune evasion in acute myeloid leukemia with CD38 CAR-engineered NK cells by Luciana Melo Garcia, Achintyan Gangadharan, Pinaki Banerjee, Ye Li, Andy G.X. Zeng, Hind Rafei, Paul Lin, Bijender Kumar, Sunil Acharya, May Daher, Luis Muniz-Feliciano, Gary M. Deyter, Gabriel Dominguez, Jeong Min Park, Francia Reyes Silva, Ana Karen Nunez Cortes, Rafet Basar, Nadima Uprety, Mayra Shanley, Mecit Kaplan, Enli Liu, Elizabeth J. Shpall, Katayoun Rezvani

    Published 2025-01-01
“…CRISPR-Cas9 deletion of CD226 led to reduced LFA-1 recruitment, poor synapse formation, and decreased NK cell anti-leukemic activity. Engineering NK cells to express a chimeric antigen receptor targeting the AML antigen CD38 (CAR38) could overcome the need for CD226 to establish strong immune synapses. …”

Carbonic anhydrase 2-derived drug-responsive domain regulates membrane-bound cytokine expression and function in engineered T cells by Mara C. Inniss, Sean G. Smith, Dan Jun Li, Benjamin Primack, Dexue Sun, Grace Y. Olinger, Kerri-Lynn Sheahan, Theresa Ross, Meghan Langley, Violet Young, Andres Alvarado, Shabnam Davoodi, Jiefei Geng, Michael Schebesta, Michelle L. Ols, Jeremy Tchaicha, Jan ter Meulen, Dhruv K. Sethi

    Published 2025-01-01
“…We develop conditional degrons using library-based screening of mutants and show characterization of one DRD using crystallography and molecular dynamics (MD) simulations. Using protein-engineering solutions to increase the valency of DRDs fused to the cargo we have developed “modulation hubs” and show tight regulation of membrane-bound cytokines IL2, IL12, IL15, IL21, IL23, and IFNα in genetically engineered T cells. …”
