Oncolytic vaccinia virus armed with anti-CD47 nanobody elicit potent antitumor effects on multiple tumor models via enhancing innate and adoptive immunity by Jie Chen, Liu Yang, Mengyuan Li, Qian Ye, Shibing Wang, Zengpeng Li, Wenbin Qian

    Published 2024-12-01
“…However, this therapeutic approach is often hampered by on-target side effects, physical barriers, and immunosuppressive tumor microenvironment (TME).Methods To improve therapeutic efficacy while minimizing toxicities, we engineered an oncolytic vaccinia virus (OVV) encoding an anti-CD47 nanobody (OVV-αCD47nb). We demonstrated the specific binding activity of αCD47nb secreted from the virus-infected cells to CD47 and that both secreted αCD47nb and OVV-αCD47nb blocked the “don’t eat me” signal of macrophages. Results Intratumorally injected OVV-αCD47nb continuously releases the αCD47nb in tumor tissues, thereby conferring superior systemic activity against breast and colon tumor cells and prolonging survival compared with OVV control. …”

Mixed radiation with different doses induces CCL17 to recruit CD8+T cell to exert anti-tumor effects in non-small cell lung cancer by Liuying Huang, Duo Wang, Muchen Xu, Danqi Qian, Yulin Cao, Xiaohan Wu, Liang Ming, Junhui Tang, Zhaohui Huang, Yuan Yin, Leyuan Zhou, Leyuan Zhou, Leyuan Zhou

    Published 2025-01-01
“…According to the findings from IHC and FCM studies, the triple therapy group exhibits a heightened infiltration of immune cells into the tumor, largely attributable to the augmented expression of CCL17 within the tumor under these irradiation regimens, which subsequently draws CD8+ T cells to infiltrate the tumor site, exerting cytotoxic effects.ConclusionOur study shows that the combined application of 20Gy and 6Gy can enhance the infiltration of tumor CD8+T cells in mice and improve the effectiveness of immunotherapy.…”

Clofarabine induces tumor cell apoptosis, GSDME-related pyroptosis, and CD8+ T-cell antitumor activity via the non-canonical P53/STING pathway by Jing Chen, Jie Wu, Xiang Chen, Qian Tao, Nian Liu, Cong Peng, Can Lu, Qiuqiu Li

    Published 2025-02-01
“…Most importantly, Clo administration dramatically increased the cytotoxic activity of CD8+ T cells in vitro and in vivo. Mechanistically, the administration of Clo induced the interaction of P53 with STING, which activated the non-canonical STING-NFκB pathway; consequently, NF-κB directly bound to the promoter regions of its target genes, including CCL5, CXCL10, HLAs and BAX. …”

The Average IFN-γ Secreting Capacity of Specific CD8+ T Cells Is Compromised While Increasing Copies of a Single T Cell Epitope Encoded by DNA Vaccine by Yanmin Wan, Jing Wang, Haizhu Zhou, Zhidong Hu, Xiaonan Ren, Jianqing Xu

    Published 2012-01-01
“…In this study, by constructing epitope-based DNA vaccines encoding a previously identified CD8+ T cell epitope, we investigated the influence of multiplying epitope copies on both the frequency and the MFI of specific IFN-γ secreting CD8+ T cells. …”
