Published 2011-01-01“…【目的】 探讨白细胞介素-10(IL-10)对弓形虫眼病小鼠视网膜组织炎症和体内γ-干扰素(IFN-γ)和肿瘤坏死因子-α(TNF-α)的影响【方法】 将120只C57BL/6小鼠分为3组:①对照组40只;②感染组(实验A组)40只;③IL-10干预组(实验B组)40只腹腔注射弓形虫感染小鼠后,观察小鼠眼部的组织学切片,对眼球和外周血做基因检测;注射IL-10后,观察其对眼部炎症反应和体内IFN-γ和TNF-α水平的影响【结果】 弓形虫感染第6天,在实验组小鼠眼部和外周血中均可检测到小鼠P30基因;房水中IFN-γ(4531.4 pg/mL)和TNF-α(1572.3 pg/mL)的浓度升高;血清中IFN-γ(456 ± 39) pg/mL和TNF-α(14.6 ± 2.2)pg/mL的浓度升高;视网膜组织可见到弓形虫,且出现明显的炎症反应;IL-10注射后,房水中IFN-γ(2482.7 pg/mL)和TNF-α(1248.1 pg/mL)的浓度降低,血清中IFN-γ(266 ± 27)pg/mL和TNF-α(12.1 ± 1.9)pg/mL的浓度降低,但仍高于对照组,视网膜组织中仍可见到弓形虫,但炎症反应减轻,将三组血清中两种细胞因子的浓度组间差异进行方差分析,P < 0.01,表明差别具有统计学意义【结论】 小鼠急性眼部弓形虫感染病变主要集中在视网膜,IL-10可减轻弓形虫感染小鼠的眼部炎症反应…”
