    “…【目的】 抵抗素(resistin)是一种与胰岛素抵抗(IR)密切相关的脂肪细胞因子65377;本研究通过观察resistin对C2C12肌细胞株葡萄糖代谢的影响,以探讨resistin在骨骼肌IR发生中的作用65377;【方法】构建含resistin基因的重组表达质粒pcDNA3.1-resistin,用于转染C2C12肌细胞株,使其表达resistin,用3H-2-脱氧葡萄糖进行葡萄糖摄取试验,用D-[14C(U)]葡萄糖进行葡萄糖氧化和糖原合成试验,RT-PCR检测肌细胞的葡萄糖转运蛋白4(GLUT4)mRMA65377; 【结果】 pcDNA3.1-resistin转染的C2C12肌细胞株可表达resistin;resistin使肌细胞胰岛素刺激的葡萄糖摄取率减少约30%,并使GLUT4mRNA 表达降低28%,而对葡萄糖氧化和糖原合成无显著影响65377; 【结论】 resistin可能通过抑制骨骼肌GLUT4mRNA的表达而降低葡萄糖摄取,但对葡萄糖氧化和糖原合成影响较小,提示resistin在骨骼肌IR发生中的作用有限65377;…”
