系统性红斑狼疮皮损角质形成细胞体外培养及细胞学观察
Published 2010-01-01“…【目的】体外培养系统性红斑狼疮(SLE)皮损部位角质形成细胞(KC),观察其细胞学特性。【方法】 原代培养SLE皮损和正常皮肤的角质形成细胞,倒置显微镜观察细胞的形态特点,免疫荧光法进行角蛋白检测并计算细胞纯度;两步消化法纯化细胞;CCK-8(cell counting Kit-8)评价细胞的增殖情况,绘制生长曲线。流式细胞仪观察细胞的生长周期和凋亡情况。【结果】使用K-SFM(serum-free keratinocyte medium)无血清培养基能成功体外培养SLE皮损的角质形成细胞,呈铺路石样。结合人工纯化,能达到 > 95%的纯度;与正常对照相比,SLE皮损角质形成细胞进入指数生长期较迟(7 d vs. 4 d),但进入后增长迅速,S期比例为: (40.68 ± 1.81)% vs. (34.43 ± 1.24)%(P < 0.05);凋亡率高:(6.08 ± 0.72)% vs. (4.32 ± 0.44)%(P < 0.05)。【结论】 SLE皮损角质形成细胞培养难度大,细胞的生物学行为存在异常,可能为SLE发病的独立、始发因素,其致病机制值得深入研究和探讨。…”
