Summary:Objective:To observe the effects of <italic>cistanche deserticola</italic> on the behavior of rat model of Parkinson's disease (PD) and expression of glycogen synthase kinase-3 beta (GSK-3β) in hippocampus, levels of β-catenin protein and its phosphorylation, and explore its mechanism in treating Parkinson's disease.Methods:Fifty male rats were randomly divided into six groups (normal group, model group, sham operation group, low dose group, middle dose group and high dose group). The rat model of Parkinson's disease was established by the intracranial injection of 6-hydroxydopamine. Behavioral step test was conducted respectively at the third, 7th and 14th day after given the decoction of <italic>cistanche deserticola</italic>. After the 14th day of behavioral testing, rats were killed to extract the protein from the hippocampus, and the expression of GSK-3β, β-catenin and p-β-catenin were detected by Western Blotting.Results:In the behavioral step test, the stride length of model group was significantly decreased compared with normal group (<italic>P</italic>&lt;0.01); after administration, the stride length of low, middle and high dose groups were increased respectively compared with model group (<italic>P</italic>&lt;0.01). In the Western Blotting, the protein expression of GSK-3β, β-catenin and p-β-catenin in model group were significantly increased compared with normal group (<italic>P</italic>&lt;0.01); compared with model group, the protein expression of GSK-3β was respectively decreased in middle and high dose group (<italic>P</italic>&lt;0.01), the protein expression of β-catenin was respectively increased in low, middle and high dose group (<italic>P</italic>&lt;0.01), and the protein expression of p-β-catenin in low, middle and high dose group was respectively decreased (<italic>P</italic>&lt;0.01, <italic>P</italic>&lt;0.05).Conclusion:<italic>Cistanche deserticola</italic> can improve the behavior disorder in Parkinson's disease, and its mechanism is probably related with inhibiting the activity of GSK-3β and expression of p-β-catenin, increasing the expression of β-catenin to start the Wnt signaling pathway and protect the PD neurons.
