| Summary: | Background: We have previously reported the addition of 10 ng/mL of granulocyte-macrophage colony-stimulating factor (GM-CSF) during in vitro maturation (IVM) increases embryo development in a mouse model of IVM. The present study determined whether 5 ng/mL of GM-CSF is a more appropriate concentration. Method: Cumulus oocyte complexes were harvested from CBAF1 female (21–23 days) mice injected with 5 IU pregnant mare chorionic gonadotropin (PMSG), and embryo development was examined following the addition of 5 or 10 ng/mL GM-CSF during IVM. Results: The addition of 5 ng/mL increased inner cell mass (ICM) cell number (13.21 ± 0.52 vs 14.60 ± 0.56). In contrast, 10 ng/mL increased cumulus expansion (1.86 ± 0.11 vs 2.34 ± 0.13), ICM cell number (13.21 ± 0.52 vs 15.54 ± 0.48) and the ratio of ICM/trophectoderm (TE) cells (40.81 ± 2.91 vs 62.46 ± 2.70). Conclusion: We conclude that 10 ng/mL is a more effective concentration for clinical trials aimed at improving IVM. |
