| Summary: | Tissue culture, including banana plants, is a very important method for plant propagation. This research aims to carry out the multiplication of local Bengkulu Ambon banana plants in vitro and to determine the best concentration of BAP Growth Regulatory Substances (ZPT) that can support the multiplication process. The method used was a Completely Randomized Design (CRD) with two factors. The first factor is the local Ambon banana genotype, namely Ambon Yellow and Ambon Green, while the second factor is the concentration of ZPT BAP, which consists of four levels, namely 0 mg/L, 3 mg/L, 5 mg/L, and 7 mg/L. This research was repeated 3 times, resulting in a total of 24 experimental units, and each experimental unit had three samples, so there were 72 plants. The results of variance analysis showed that the Genotype and BAP treatments affected the variables of plant height, number of leaves and leaf length but had no effect on the number of shoots. The BAP treatment affected all the variables observed: plant height, number of leaves, leaf length and number of shoots. Interactions also influence all observed variables. The interaction results showed that the 5 mg/L BAP treatment was best for the growth of the Green Ambon genotype because it could increase plant height growth by 11.6 cm, shoot number by 15.7 shoots, increase the number of leaves by 6.43 pieces, and increase leaf length by 8.86 cm. Meanwhile, BAP treatment of 7 mg/L was best for the growth of the Ambon Yellow genotype because it could increase the growth of plant height by 12.93 cm, number of shoots by 15.93, number of leaves by 11.33 cm and leaf length by 10.96 cm. Shoot multiplication in vitro was influenced by external and internal factors. |
